Biosynthesis of liver membranes. Incorporation of ( 3 H)leucine into proteins and of ( 14 C)glucosamine into proteins and lipids of liver microsomal and plasma-membrane fractions.

چکیده
1. The smooth-and rough-microsomal and the light and heavy plasma-membrane fractions of mouse liver homogenates were prepared and characterized by using biochemical markers. 2. The hexosamine/protein ratio was threefold higher in the plasma membranes than in the smooth-microsomal fraction. Glucosamine was bound only to protein, and galactosamine was attached mainly to lipids. 3. [(3)H]-Leucine and [(14)C]glucosamine were injected into animals and the rates of incorporation of radioactivity into the fractions were determined. Both precursors were rapidly incorporated into the microsomal fractions, but plasma membranes showed a slower rate of synthesis which reached a maximum at 2-4h after intravenous administration. 4. The light- and heavy-plasma-membrane fractions showed similar patterns of incorporation, and therefore a precursor-product relationship appears unlikely. 5. Plasma membranes, especially the light subfraction, showed appreciable incorporation of hexosamine into chloroform-methanol-soluble components which were shown to be mainly glycolipids. 6. The results indicate that liver plasma-membrane proteins and glycoproteins are synthesized at similar rates. However, glycolipid synthesis in plasma membranes occurred more rapidly.

Characterization of biophysical properties of single chloride channel in rat brain mitochondrial inner membrane by channel incorporation into bilayer lipid membrane
Introduction: Recent studies have shown the presence of Cl- channels in heart and liver mitochondrial membranes. In this work, we have characterized the functional profile of a Cl- channel from rat brain mitochondria. Methods: After removing and homogenizing the rat brain, the supernatant was separately centrifuged in MSEdigitonin, H2O and Na2CO3 and mitochondrial inner membrane vesicles wer...

nL-leucine-l-C 14 was administered by intracardiac injection to guinea pigs and its in vivo incorporation into the proteins of various pancreatic cell fractions followed over a period of 2 hours. The pancreas was homogenized in 0.88 u sucrose and fractionated by differential centrifugation to give nuclear, zymogen, mitochondrial, microsomai, postmicrosomai, and final supernatant fractions. The ...

متن کاملBiosynthesis of plasma lipoproteins. Incorporation of 14C-glucosamine by cells and subcellular fractions of rat liver.
The incorporation of glucosamlneJ4C into plasma lipoproteins has been measured in the intact rat, in liver slices, and in the liver microsome fraction. The labeling of lipoprotein was studied in microsome subfractions, both in viva with glucosammeJ4C and in vitro with UDP-N-acetylglucosamineW. After isolation of lipoproteins with the aid of carrier plasma, labeled lipoproteins were found in smo...
